The in vitro transcription units of bacteriophage φX174. III. Initiation with specific 5′ end oligonucleotides of in vitro φX174 RNA

Abstract

Four major 5′ end oligonucleotide sequences are detected in 5′ proximally labeled in vitro bacteriophage φX174 transcripts. These are correlated to the three major initiation sites previously identified by preparative hybridization and nucleotide fingerprint analysis. In addition, the 5′ end oligonucleotides of the several discrete components isolated by preparative slab gel electrophoresis, also 5′ proximally labeled, are identified. The results confirm the genetic locations of the three major transcriptive initiation sites previously postulated.
