The Process of Infection with Bacteriophage φX174: VI. Inactivation of Infected Complexes by Ultraviolet Irradiation

Abstract

The sensitivities to ultraviolet irradiation of complexes formed by infection with bacteriophage φX174 in Escherichia coli C, E. coli C_s (a strain lacking the capacity to perform host cell reactivation), and E. coli CR (a strain requiring thymine) were determined at various wavelengths, at various stages of development, and under various growth conditions. Neither irradiated bacteriophage φX174 nor irradiated free φX DNA is subject to host cell reactivation; however, irradiated replicative form φX DNA is. The target that is observed in the ultraviolet inactivation curves of the complex after three minutes of infection is also subject to host cell reactivation. In the absence of host cell reactivation, the ultraviolet inactivation curves suggest the existence of either one or a very few sensitive targets in the infected cell.
