Secondary isotope effects in dehydration of malic acid by fumarate hydratase

Abstract
The kinetic isotope effects for the dehydration by fumarate hydratase of 2-^3H-1,4-^(14)Cl-/-malic acid and 2,3-^3H-1,4-^(14)C-l-malic acid have been determined and support a carbonium ion mechanism for the enzymatic reaction. The isotope effect on the equilibrium between l-malate and fumarate has also been determined experimentally and agrees closely with that anticipated on the basis of a Bigeleisen treatment.
