The in vitro transcription units of bacteriophage φX174. II. In vitro initiation sites of φX174 transcription

Abstract

Employing the 5′ proximal labeling procedure reported previously, and hybridization to Haemophilus endonuclease bacteriophage φX RF DNA fragments, we have detected three major sites of initiation of in vitro φX transcripts. These three sites have been located on the φX174 genetic map in the following regions: (1) near the 5′ end of gene A, (2) near the 5′ end of gene B, and (3) near the 5′ end of gene D.

Espejo for his gift of ^3H-labeled φX DNA and his suggestion to use it as an internal hybridization control. We also wish to thank Drs P.H. Johnson and A. S. Lee for their generous gift of Haemophilus endonuclease preparations. This research was supported in part by grant GM13554 from the National Institutes of Health.
